I think it would depend on who your current situation is. Do you have that many minor leaguers that you need to protect from losing, then I would fill it up. If you have minor leaguers who you don't mind losing, that aren't prospects then maybe it's worth taking a chance on someone.

Just put on minor leaguers you think have a chance of making your team. Don't wast spots on career minor leaguers. Remember also if your 40 man is full you cannot participate in the rule V draft.

If your in contention, yes, it should be filled up (atleast 39) as you should have solid depth, and to prevent against injuries.
If your rebuilding, Id keep it 35 or so, so you can add Rule V eligbile players, participate in Rule V draft, and chances are your going to be moving guys up and down, using waiver players ect ect. It all depends on your teams situation. for example, in my solo league (Trans-Atlantic Baseball Association, dynasty forum....enough shameless self promotion), I've had mine nearly full all year (give it a few guys had been released but others have immediately taken their spots), since I dont have many Rule V eligible guys, and I need depth (and have played musical chairs with my bullpen and 5th starter, including picking up 3 waiver wire guys to fill those spots). However, if I was rebuilding and mixing in and out new lineups every few months, id try and keep it as empty as possible so as to provide me with the most flexibility as possible in adding players and subtracitng them from the 40 man roster). Of course, by the off-season (im in september so its full of callups now) I plan on trimming it down to the 32 range for transactions, but filling it back up by the start of June (I have a ton of interchangeable fring players at 3B-RP-5th starter-backup OF). So I suppose, in the end, it comes down to how cement your roster is. If you have a nearly set roster, then go ahead and fill it up (leaving maybe a slot open), but if your constantly calling up/demoting/cutting/picking up/signing guys then your gonna wanna keep it with quite a few empty slots

I would only add players to the 40-man if you absolutely have to to protect them, etc. You can always add players later if you find that you need to. Plus, it's much easier to add players later than it is to remove players if you find that you've made a mistake in who to include.

Here's a tip... When I imported from 5 to 6, I Started with 25 men on the 40 man roster. They were the 25 men on my active roster and I add them through the year as the situation is needed. I have not made it to the Rule V draft yet, however, when I do I will add any players that are unprotected that I would like to keep. Hopefully it isn't more then 40.

In MLB I don't think it is required that they keep the 40 man full.
