Lineup vs. Depth chart - Position
It seems I can set the position of a player in either the lineup or on the depth chart. Does one override the other? OOTP seems to let me setup a player as 1B on my lineup and DH on my depth chart.

Off the top of my head, I'd have to say depth charts offer you the flexibility to have a player back up nearly any position. The lineup sets his position in the field. If you look at the depth charts' starter column, I don't think you can find one different than the one in the lineup. Does that make sense? The starter column, for me, took awhile to get used to reading as it doesn't line up with the lineup.

I believe the lineup overrules depth chart, so if you have somebody starting at one position and backing up another, he will be unavailable for the latter.

So, if lineup overrides depth chart then why even have a starter section in the depth chart? It seems like the starter section of the depth chart affects nothing in the game.
