One Human Manager with multiple teams

Is it possible for the same human manager to function as such with more than one team?

I have made us both commissioners so we can do what we need with our teams but I am the only commissioner who is sim-ing the game and updating the league file. The managers have different names but have commissioner access. So one human is manager "Mike B" and "Michael W."

However, my other human manager is saying he cannot download the latest league file as it does not exist. Could it have anything to do with my human manager having two teams?

Yes that is fine. You can control as many teams as you wish.
Check your firewall if you cannot download the file.
