When does the game no longer consider a prospect a prospect?

Is it as soon as they make their Major league debut? What if they only play a handful of games before being sent back down to the minors?

A prospect is any player whose rights are owned by a professional team, but who has yet to surpass a threshold where they achieve rookie status (as defined by their respective league), or is not established with the team yet.

That means service time for MLB.

The Player Development tab mentions 75 days as the cut off. I'm not sure that is correct though because very recently I happened to be monitoring the ranking of my farm system very closely trying to achieve an owner goal and I noticed that my top guy suddenly dropped out of my prospect list and he only had between 40 and 50 days of MLB service time (forget the exact number). Cost me 3 spots in the ranking grrr.

I think I've seen 45 days somewhere - which matches the rule for rookie eligibility.

I had a player on the top prospect list with exactly 74 days' service time, which probably means the cutoff is still 75.

IRL in MLB a player loses rookie status when they reach 130 ABs or 50 innings, or they have 45 days on the active roster. I am not sure that OOTP reflects the 45 days on the active roster, but I would think it goes by the ABs and IP. You probably lose prospect status when a player is no longer a rookie.
