Some of the basics about the 40-man roster:

-- A player has to be on the 40-man roster to be active in the majors

-- Players on the 40-man roster cannot be selected in the Rule V draft

-- Once a player is on the 40-man roster, they cannot be removed from it without one of two things happening:

* Waivers -- A player who is placed on waivers can be removed from the 40-man roster.

* Designated for assignment -- If you DFA a player, they are immediately removed from your 40-man (I think). You then have 14 days to either trade them or pass them through waivers. (If they clear waivers, they can then be assigned to the minors, or released.)

In real life, a team's 40-man roster is usually composed of their MLB regulars, minor leaguers whom the team expects to shuttle up and down between AAA and the parent team, and high value prospects that the team wants to protect from the Rule V.

For OOTP, players acquired by trade are not automatically placed on your 40-man roster. They appear in your DFA pool (which, although it might seem counterintuitive, is actually correct, since the acquired player needs to be assigned to a team). If you want to activate them, you need to have a 40-man roster spot available, or make one available by DFAing/waiving a player.
