You would need to start a new game if you really care about college players getting the ratings boost from this update, as your existing save will have a few draft classes already formed and those college players will not have their ratings boosted.

Most of the changes to college players is actually related to their development while in college, not necessarily as much the initial roster ratings for them. So even that point you wouldn't necessarily need to start a new game to catch most of the updates (other than obviously the current draft players have all been significantly updated since the draft itself).
