Anyone in the organization is eligible as a replacement.
So basically the rule right now is:
-anyone on the 40-man on Aug 31st is immediately eligible
-anyone on the DL on Aug 31 is eligible
-anyone in the organization can be made eligible if they replace someone who is on the DL.
You can swap out 40 for 25 in the above case depending what option in OOTP you're talking about/what MLB rules were a few years ago.
I do believe that we handle the DL/replacement players ever so slightly differently than the actual MLB rules, but very close.
