Having "ALL" players from that era available would not be do-able. But since you are open to using fictional players, if you use Spritze's db, turn off both Retire According to History and Players Miss Seasons Accdg to History, you should be able to have something pretty cool and close to what you want.

As an example, I started a historical league in 1890, pretty much the basic default OOTP historical setup. I left it with just the 16 MLB teams - with reserve rosters - for the first 12 years. Then, for 1903, I added a PCL and Federal League - both of which combine to serve as the Triple A affiliates for MLB, and I also added an independent Union Association (6 teams). I'm currently in 1908 and have had very little trouble stocking teams with players. Once in awhile I run a little short of position players (not catchers) on the Free Agent list, but I I can offset this by releasing a few players from some Triple A teams that are overstocked... And besides, I'm not using any fictional players; since you would be using fictionals, you should have no problem.

So if you were to start earlier - maybe 1890 or something like that - and let the game run (in order to build up reserve roster depth) before you get more involved and add your PCL & SA, your idea should work pretty dang close to what you want.

NOTE: I doubt you'd want to but DO NOT use real historical transactions unless you use the odb file from v12; v14 will not work with what you want to do... I don't know much about the v13 odb transactions file but I don't think it would work for you, either.
