There are two ways to do it, but both require the player clearing waivers if you want to keep them.
Waive them first, see if they clear waivers, then remove them from the 40 (need to demote them to the minors if they're on the major league roster).
or
Waive and designate them for assignment at the same time. This immediately frees up the 40-man spot, then you wait to see if they clear. When/if they clear, then you demote them to the minor league level of choice.
