Programs that are capable from running off a flash drive, I believe, need to have all the files and parameters stored locally within its own sub-directories.
The program cannot use the registry or other locations on the install computer (such as My Documents or Local Settings) because that essentially means that when you move your flash drive from the first PC to a different PC, you are not carrying those critical files and parameters with you.
That said, OOTP uses the registry, especially for the eLicense information, as well as storing a great deal of its files outside of the parent directory of the executable.
So in short, you may be able to install it on a flash drive, but it's only going to work on the one computer that you install it on.
