It's a daunting task to check every lineup and pitching rotation. If this is just your solo league try:

1. Make a backup of your league and tuck it away in a folder somewhere other than inside the OOTP6 folder.

2. Fire up OOTP6

3. Go to the League Setup page and then click "Run computer manager on all teams" This should take care of lineups and pitching rotations.

4. Now try to run something. If everything o.k. now, Steve would be right. If not, try disabling any security or anti-virus software. They frequently cause problems for the game. If you are paranoid about security, temporariliy disconnect your pc from the internet.
